The congregation was gathered in 1654, when Flatlands was the Dutch town of Nieuw Amersfoort, and the first church on this ground was an octagonal shingled building. The Greek Revival church standing at Kings Highway and East 40th Street replaced it in 1848. The burial yard beside it holds roughly fifteen hundred graves, the earliest from 1660.
